Purpose: To investigate self-made freeze-dried amniotic membrane's effect of keeping the filtrating pathways open in the glaucomatous filtrating surgeries in rabbit eyes.Methods: We prepared the healthy human amniotic membrane (AM) according to the routine reference, freeze dried after depriving the epithelial cells, then vacuum packed, stored for 2 months at the room temperature. The resultant freeze-dried AM was then hydrated,and characterized for its physical, biological, morphologic properties by stretch stress tests, HE staining, light microscopy and electron microscopy. In addition, trabeculectomy were performed on 38 eyes of 24 rabbits which were divided into 3 groups: group A (9 rabbits)was treated with the freeze-dried AM under sclerotic flap, group B (5 rabbits) was performed trabeculectomy using MMC (0.25mg/ml) under sclerotic flap and group C (5 rabbits) played the role of control which had surgery only. Anti-inflammation eye drops were given for each group's rabbits. 4 rabbits were selected randomly from the group A for the pathologic examination, one rabbit was killed from them in 1,2,3,4 weeks after operation and extirpated the eyeball to observe the decomposition of the amniotic membrane, fibroblast and inflammatory cells by HE staining. Other rabbits'postoperative examinations including the filtering bleb, reaction of the anterior chamber and complications everyday, and in 1,2,3,7,14,21,28d the intraocular pressure(IOP)were measured and noted.Results: After hydrated, the self-made freeze-dried AM had the good transparency and tenacity, and the stretch stress test revealed no significant differences between freeze-dried AM and fresh AM, it's physical property...
