| Objective:In order to find the factor improving the development and implantation of embryo in vitro by decting the secreting of leptin in different culture conditions.To determine between"leptin"or"blastocyst score"and"implantation and pregnancy outcome"by use mouse embryo transplantation model ,and then, find out the comprehensive factor to evaluate embryo quality .Method: 2-cell embryos were cultured to 4-cell,8-cell,morulas and blastocysts stage respectively in three different culture systems:(1)simple sequential culture group;(2) coculture and sequential culture group with human endometrium epithelial cells;(3)natural development in mouse reproductive tract. These embryos continued to be cultured for 6 hours in the 50ul culture medium microdrops separately,and then,we removed the embryos,collected these culture microdrops and detected the secretion of leptin by ELISA method.The microdrops were stored at -20℃for detecting of leptin..Those of the removed embryos in natural development,having developed to blastocysts and getting a pre-implantation score of 3AA or 3BB,were transferred into the uterus of pseudopregnancy mice on the third day.11days later ,we observed the implantation rates and compare the effect of the secretion of leptin and blastocyst score on embryo imbedding ability.Results: 1.The embryo growth rate ,after 8-cell stage ,and blastocyst formation rate in coculture and sequential culture group were significantly higher than that in simple sequential culture group(P<0.05). 2.We couldn't detected the expression of leptin in the culture microdrops in different growth phases from 2-cells embryos to 8-cells embryos in three different... |
