| Objective:To observe the effect of agent ZS on selenite-induced cataractous lenses of rats.Methods:1. Preventive effect: 26 Wistar rat pups(day 8~10 post partum) were divided into three groups: normal control,model,protective groups. The normal group received the normal saline (0.1ml/10g b.w.) by intraperitoneal injection daily for 6 days. The model group received sodium selenite (30μmol/kg b.w.) intraperitoneal injection only once. The protective group received sodium selenite(30μmol/kg b.w.) intraperitoneal injection followed by agent ZS (20μmol/kg b.w.) intraperitoneal injection in 30 minutes in the first day and then received agent ZS daily for 5 consecutive days. At day 7 the transparency of all lenses were observed through slit lamp microscopy. At the same time, the levels of Superoxide dismutase(SOD),Malonaldehyde( MDA)were measured respectively.2. Therapeutical effect: 55 Wistar rat pups (day 8~10 post partum) received subcutaneously sodium selenite (30μmol/kg b.w.). The rats with selenite induced cataract were divided into two groups: grade IV and gradeV,according to the degree of the lens opacity. Then the two groups were divided respectively into three subgroups: model,treated for 1 month group and treated for 3 months groups. In the grade IV group, the model group was administered daily by intraperitoneal injection normal saline (0.1ml/10g b.w.) for 3 months; the treated for 1 month group was administered daily by intraperitoneal injection agent ZS (20μmol/kg b.w.) for 1 month; the treated for 3 month group was administered daily by intraperitoneal injection agent ZS (20μmol/kg b.w.) for 3 month. In the grade V group, the model group was administered daily by intraperitoneal injection normal saline (0.1ml/10g b.w.) for 3 months. The treated for 1 month were divided into three groups: eyedrop application,low dose and high dose groups. The eyedrop application group was administered twice daily by eyedrop application 10mmol/L agent ZS for 1 months ; the low dose group was administered daily by intraperitoneal injection agent ZS (20μmol/kg b.w.)... |
