It is now firmly established that the growth and metastasis of the vast majority of neoplasms are dependent on the formation of adequate vasculature, i.e., angiogenesis. Endothelial cells play an integral role in a variety of physiological and pathological processes and endothelial cells play a critical role in tumor progression and metastasis by forming capillary networks that encourage tumor growth and by promoting the attachment of circulating tumor cells to the vascular wall of distant tissues. The study of these phenomena has been greatly enhanced by the development of tissue culture techniques. From these investigations, an important concept has emerged, namely, that endothelial cells are heterogeneous cells with distinct structure and function within different regions of the vasculature. The isolation and propagation of endothelial cells derived from human primary hepatic cancer, consequently, are important prerequisites to the in vitro study of primary hepatic cancer. So we explore the methods of culturing primary hepatic cancer -derived vascular endothelial cells(VEC) for its verification and phenotype study.Methods1. Trypsin and collagenase enzyme method, tissue nubbles cultivation and vascular fragments cultivation were used to isolate vascular endothelial cells, then observe which was the best one for the isolation and propagation of endothelial cells derived from human primary hepatic cancer.2. Vascular endothelial cells derived from primary hepatic cancer were identified by human factorⅧ-related antigen observed by immunocytochemistry and Weibel—Palade body detected by electronmicroscope.3.
