ObjectiveTo investigate the possible mechanism of basil polysaccharide in inhibiting tumor metastasis. MethodsHuman breast cancer cell lines MDA-MB-435 and MDA-MB-231 were cultured with basil polysaccharide under normoxia (20%O2,5%CO2) and hypoxia (1%O2, 5%CO2, 94%N2) respectively. Cell proliferations was evaluated by MTT assay; cell invasivenesses was tested by Matrigel method; expression of mRNA and protein levels of osteopontin (OPN) were determined by RT-PCR and ELISA, respectively; Gene expressions of MDA-MB-435 in hypoxia were examined by gene chip. ResultsThe 50% inhibition concentration (IC50) of basil polysaccharide for human breast cancer cell lines MDA-MB-435 and MDA-MB-231 in normoxia was 100.71ug/ml and 143.69ug/ml, respectively, and the IC50 of basil polysaccharide for MDA-MB-435 and MDA-MB-231 in hypoxia was 119. 66ug/ml and 258. lug/ml, respectively; the number of invasive and migrative cells declined and the expression of mRNA and protein levels of OPN declined when treated with basil polysaccharide(P<0.05); genes for Aldolase, phosphoglyceromutase, Ldh, Agxt, Ctsk, G6pc , Hyase, et al were up-regulated and genes for MHO-I , Cd81 , et al were down-regulated with...
