Objectives: The culture method was modified in order to get pure and more osteoblasts to study bone diseases. In this study, the BuShen Chinese medicine was administered to the osteoblasts to investigate its biological effects. Methods: The tissue pasting and enzyme digesting were combined to obtain osteoblasts. The osteoblasts were observed under convert microscope. The cells were identified by cellular morphology, alkaline phosphatase (ALP) dying, calcium nodule formation, osteocalcin immunochemistry and immunofluore-scence of collagen type I. 10% rat scrum contained different concentration BuShen Chinese medicine were added into culture medium for 72h as the study groups, and 10% rat serum contained no BuShen Chinese medicine as the control. The flow cytomety, bone ALP, osteocalcin, 3H -TdR mixing and RT-PCR were used to observe the effect on the proliferation and endocellular Ca2+ concentration of osteoblasts. Results: 1. The cells cultured were positive for ALP dying, and calcium nodule were formed, and they were black with the Von-Kossa dying.
