Objective: 1. To build the model of Trabecular Meshwork Cells in vitro . 2.To research the effect mechanism of serum with Chinese medicine component to Trabecular Meshwork Cells cultrured in vitro. 3.To investigate whether the effect mechanism of serum with Chinese medicine component can reduce the concentrations of hyaluronic aci(HA),laminin(LN)and proteincollagenâ…¢(PCâ…¢). 4.To find a new way for researching glaucoma in vitro.Methods: 1.Forty-eight 3 months old SD-rats were feed with different concentration QingGuangAnKeLiJi and YiMeiKang Tabelt for getting different concentration serum with Chinese medicine component. 2. To testify the fingerprint geography of serum with Chinese medicine component, we choose HPLC technology to compare the component of liquid involing medicine with the countpart of serum with Chinese medicine. 3.Put different serum with Chinese medicine component to Trabecular Meshwork Cells cultrured in vitro within 24 hours, after another 24 hours, Trabecular Meshwork Cells were collected and the sum of Bcl-2,Bax were tested by Flow Cytometry. 4.The different serum with Chinese medicine component was added to bovine trabecular meshwork cells 3th subcultured in vitro After 24 hours,radio immunity was used to analyze the concentrations of HA, LN and PCâ…¢ Results: 1.Successfully get the serum with Chinese medicine component and using for Trabecular Meshwork cells cultured in vitro. 2.The HPLC of the serum with Chinese medicine differeciates with countpart without Chinese medicine obviously. And it not only has the same peak, but also has some difference with the liquid with medicine. Though the concentration of serum with Chinese medicine varied,the fingerprint geography have the same peak, the square of each peak is not equality. 3. The mixed cultured Trabecular Meshwork Cells were effected with different serum with Chinese medicine component. The expression of Bcl-2 increased as the concentration of serum with Chinese medicine component increased, but the expression of Bax decreased as he concentration of serum with Chinese medicine component increased. The group of QingGuangAn-20 was superior to the control group and the model group(P<0.01 ),and the group of QingGuangAn-20 was superior tothe group of YiMeiKang-5 (P<0.05) ; the group of QingGuangAn-10 has no distinct difference from the group of QingGuangAn-5 and the group of YiMeiKang-5. 4.in the different serum with Chinese medicine component groups , the concentration of PCâ…¢, HA and LN was simultaneously reduced.Conclusions: 1.It concluds that the concentration of medicine dosing will influence the metabolism concentration caused by QingGuang -AnKeLiJi.The method is simple and with a good reproducibility. 2.The serum of QingGuangAnKeLiJi can postpone the expression of apoptosis and pretect the trabecular meshwork cells in vitro. 3.The serum of QingGuangAnKeLiJi can reduce the concentrations of hyaluronic acid ,laminin and protein collagenâ…¢.
