Research On The Mechanical Function Of Reinforcing Qi And Activating Blood Of Astragalus And Angelica

Astragalus membranaceus combined with Angelica sinensis (A.A for short) is one of famous herb couples, it has different effect with different combinational ratio.This task is based on the results of the former, selecting the fifth and the sixth group as objects, the former had better effect on the platelet aggregation rate of acute blood stagnation in rat and the latter on the whole blood viscosity, to further investigate the mechanical function of Reinforcing Qi and Activating Blood of A.A. Using the method of hunger and fatigue to establish Qi-Deficiency and Blood-Stasis model rats, the content of NO in blood plasma was measured with enzymeimmunoassay method, the level of NO markably raising in model rats was detected. Both large and small doses of A.A in the ratio of 3 to 1 and 3 to 2 can significantly reduce the content and regulate the function of blood vessel(P<0.01 or 0.05). The expression of Platelet Endothelial Cell Adhesion Molecule(PECAM-1/CD31) and P-Selectin(CD62p) on platelets membrane was determined by flow cytometry, all of doses can effectively decrease the excessive expression of CD31 and CD62p(P<0.01 or 0.05); CD54 expression on the surface cells of endothelial vessels of model rats were investigated by using immunocytochemical staining technique, obvious the high lever of CD54 expression on the surface cells of endothelial vessels and endotheliocytes of hepatic sinusoid of model rats was observed, both large doses of A.A in 2 different ratio can low the former(P<0.05), only large dose of A.A in 3:1 ratio can effectively reduce the latter(P<0.05). So it was concluded that decreasing the content of NO in blood plasma, inhibiting the excessive expression of CD31,CD62p on platelet membrane and CD54 in endothelial vessels, hepatic sinusoid may be the molecular mechanism of Replenishing Qi and Promoting Blood Circulation.
