Objective:To analyze the clinical medication rule of Professor赵泉霖in the treatment of diabetic cardiomyopathy,to dig out the common core drug pair of Astragalus and angelica,to verify the effectiveness of the drug for the prote-ction of diabetic cardiomyocytes through the in vivo experiment,to further explore the mechanism of its effective active ingredients in the intervention of high glucose induced myocardial inflammatory injury through the in vitro experiment,at the same time,through the gene sequencing data Mining and predi-cting the potential regulatory mi RNAs will provide a new way to further explore the concept of prevention and treatment of the disease and explore new drugs.Methods:1.By collecting the relevant prescriptions and drugs of the tutor-s in the outpatient follow-up and ward electronic medical record system for the treatment of diabetic cardiomyopathy,the relevant medication laws were calculated,and the key drug pairs were found out.2.Verify the effective effect of key drugs on the general situation,blood lipid and blood glucose,echocardiography and other aspects of diabetic mice from the invivo experiment,and further select drugs to carry out in vitro experiments on the best active ingre-dients astragaloside,ferulic acid,in order to further explore the interven-tion of key drugs on NF-κB related inflammatory factors and their signal pathways.3.Sequence the genes of each group of myocardial cells under the inte-rvention of high glucose in vitro,analyze and mine the data,combine the moder-n bioinformatics analysis method,carry out the KEGG enrichment analysis and go analysis of the gene chip information,and predict the high differential expression mi RNA which is most closely related to the inflammatory response.Results: 1.A total of 277 effective prescriptions were collected through the electronic medical record system of outpatient service and ward,among which the most frequently used drugs were those for invigorating qi and promoting blood circulation,and those for promoting blood circulation and removing bloo-d stasis.At the same time,statistics were made on the sexual taste and meridi-ans of traditional Chinese medicines used in the clinical treatment of diabetic cardiomyopathy.2.Through in vivo experimental modeling and intervention in the traditional Chinese medicine group and the metformin group,it was found that astragalus angelica could improve the general situation of the model mice,key indicators of blood glucose and lipid,echocardiography of the heart blood flow,as well as the morphological changes of the brain,which proved the effectiveness of astragalus angelica in improving diabetic cardiomyopathy.3.Throu-gh the in vitro experiment selected Chinese medicine group,model group,met formin group the best intervention time(72h),the best concentration(astragal-us membranaceus methyl glucoside:100umol/l,ferulic acid:20umol/l,metformin:200 umol/l),Chinese medicine group best ratio(1:1),ELISA and Western blot detection related indicators,known:drug intervention group can obviously reduce the NF-κB pathway related indicators(NF-κB,IL-6,TNF-α,MCP-1)expression.4.Gene chip detection and enrichment analysis of differentially expressed genes were performed to obtain significant mi RNA in 14 differentially expre-ssed groups.At the same time,the target gene mirna-4508,which is most closely related to NF-κB signal pathway,was predicted by further bioinformatics mining,suggesting that it may be related to the NF-κB pathway that interferes with the inflammatory response of diabetic cardiomyopathy.Conclusion:1.The pathological attribute of diabetic cardiomyopathy is based on deficiency and excess,based on deficiency of Qi and blood stasis.The treatment should be based on the principle of Invigorating Qi and activating blood circulation.2.Astragalus membranaceus is the most commonly used medicine in clinical treatment of diabetic cardiomyopathy.Most of the drugs are bitter,pungent and sweet.Most of the drugs are warm,which belong to liver,spleen and heart meridians.The treatment should be based on tonifying deficiency,promoting blood circulation and removing stasis.Astragalus membranaceus and Angelica are the most commonly used representative drugs in clinical treatment.3.Astragalus angelica and its main active components astragaloside and ferulic acid can play an anti-inflammatory role in protecting cardiomyocytes by regulating NF-κB pathway.The mechanism may be related to mirna-4508. |
