Objective:To study the possible injury mechanism in the rat aortic endothelial cells induced by DL-homocysteine in vitro and observe the interactive profect effects of folic acid, VitB6, VitB12 to RAEC injuried by DL-homocysteine.Methods:1. Observe the detriment of DL-homocysteine to RAEC by oxidative stress and (or ) decreasing the bioavailability of NO . Rat aortic endothelial cells were cultured and inoculate in 96 cells in vitro . Cell counting of detached cells were performed with a hematocytometer . the cell survival was monitored bt MTT assay. And exposed to different concentrations of DL-homocysteine ( 0.05,0.1,0.5,1.0,5.0)mmol/L, subgrouped by randomized blocks. Observe the influence of Hcy on the content of cellular MDA, NO and the changes in the activity of major antioxidant enzymes (SOD, GSH-PX) by repeated measurement design at 6h, 12h, 24h .2. Effect of DL-homocysteine on the expression of VCAM-1 and the adhesion between endothelial cells and leukocytes in cultured rat aortic endothelial cells . DL-homocysteine were subgrouped to four concentrations and a control group. Observe the difference of intensity of light of RAEC induced by DL-homocysteine for 24h and the comparison of VCAM-1 mRNA's level . Inoculate rat aortic endothelial cell with histoleucocyte for 24h , compare the rate of cell adhesion bt OD value of coomassie brilliant blue.3. The interactive profect effects of folic acid, VitB6, VitB12 to RAEC injuried by DL-homocysteine. Observe the protect effects of folic acid, VitB6, VitB12 to RAEC...
