Objective: (1),To test the hypothesis that autogeneic bone marrow stromal cells (BMSCs) grafted intravenously into the paraplegic rabbits can migrate to the impaired region,survive there,differentiate into neuron-like cells and promote restoration of structure and neurological function of the injured spinal cord . (2),To study the effect of BMSCs grafted intravenously and followed by brain derived neurotrophic factor (BDNF) infused intrathecally on morphological repairing and functional recovery of the traumatic paraphegia of spinal cord in rabbit.Methods: BMSCs were aspirated from Newzealand rabbits weighting 2.0~2.5kg and cultured in vitro . To detect the surface marker antigens with FACScan flow cytomete (FCM) when the BMSCs proliaferaterd into the fourth generation . The models of rabbit's traumatic paraplegia were made through modified Allen punch method . The rabbits were devided into five groups randomly including control group,BMSCs group,BDNF group,BMSCs+BDNF grorp and pseudo-operation group . After the models were made successfully 3 days later, the rabbits of control group were injected NS 1ml respectively ; BMSCs group's were injected BMSCs(2×106) labelled by DAPI; BDNF was infused intrathecally in BDNF group's; The BMSCs+BDNF grorp's were injected BMSCs intravenously(2×106) and followed by BDNF infused intrathecally.Recovery of conductive function of rabbits was determined by the techniques of cortical somatosensory evoked potential (CSEP),modified Tarlov score and horseradish peroxidase (HRP) retrograde tracing after infusion 1,3,5W later.To detect the survival,migration,differentiation of BMSCs in spinal cord with fluorescence immunohisto-chemistry.Results: BMSCs can adhere the wall and have polymorphic characteristic in vitro.
