| ObjectiveAngiogenesis is fundamental process by which new blood vessels are formed. It is essential in the development, invasion and metastasis of tumors. There has been many methods of three-dimensional endothelial cell culture, which provide a new thoughtway for the methods of tumor-therapy. The intention of this research is to study the difference of two methods of three-dimensional endothelial cell culture in vitro and to provide experimental support for the discovery of new ways of tumor-therapy.Methods1.Fill trypsin-EDTA into human umbilical vein to obtain sing-cell mixing suspension.2.Plate cells to the cultivate plate repeatingly to obtain purified HUVECs.3.Identifying HUVECs by the technique of immunohistochemistry as CD31,CD34 and factorâ…§is reference.4.In vitro three-dimensional culture of endothelial cell.4.1 In vitro three-dimensional cluture of endothelial cell in the surface culture system.4.2 In vitro three-dimensional culture of endothelial cell in the mixing culture system.5.Comparison of two methods of three-dimensional endothelial cell culture by statistic analysis of the experimental results.Result1.Single-cell mixing suspension most of which are HUVECs is obtained by filling trypsin-EDTA into human umbilical vein.2.HUVECs at the purity of 99.5% are obtained by plating cells to the... |
