| [Objective]To investigate the curative effect of the transplantation of mesenchymal stem cells (MSC) in the course of hepatic fibrogenesis and hepatocirrhosis in rats. [Methods]Combined with stationary culture the mesenchymal stem cells of male SD rat were isolated and purified by density gradient centrifugation. Then the cells were expanded by subculture and the forth passage cells (5×106ml), proven to be capable of differentiation into osteoblasts and lipocyte, were respectively injected into the homogeneous female rats via the caudalis vena on the fist day, the sixth week and the eighth week of the hepatic fibrosis model induced by carbon tetrachloride ( CCL4 ) in rats. Four weeks after MSC transplantation, the pathology situation of liver in the transplantation group and the control group were evaluated with Scheuer inflammation and Schmid M fibrosis activity grading methods. The expression of Y chromosome in rat liver was also tested by the method of fluorescence in situ hybridization. (FISH). [Results]6 out of 12 rats from 1st day transplantation group were rated less than 1 point using Scheuer inflammation activity grade, while only one in the control group had similar point The mean value of Scheuer inflammation score in transplantation group was 1.50, and 2.55 in the control group (t=2.894, P=0.009). The mean value of Schmid M Score in transplantation group was 0.75, and 0.82 in the control group (t=2.894, P=0.009).4 out of 12 rats from 6th week MSC transplantation group scored 4 points and 3 scored 6 points using Scheuer inflammation activity grading method. The corresponding number of samples with such scores in the control group was 1 and 7, respectively.
