Pharmacokinetics Of Yi-Tang-Kang Capsule In Rabbit

Yi-Tang-Kang is a new king of traditional Chinese medicine for diabetes.It is compacted by twelve Chinese medicinal materials contain ginseng radix rubric Coptis chinensis cortex phellodendri pueraria radix et rhizoma rhei fructus schizandrae, Panax pseudo-ginseng etc.Yi-Tang-Kang can prevent and cure diabetes and diabetes'complication by kata- amylaceum kata- lipin anti-damage of DNA.It's effect of kata- amylaceum kata- lipin anti-damage of DNA can reduce disease incidence,save money for diabetes,it have a huge society and economic retums.There is no same report about Yi-Tang-Kang's pharmacokinetics.Objective:The purpose of this study is to establish a quantitative measurement for berberine hydrochloride in cony blood plasma after feed with Yi-Tang-Kang and the pharmacokinetics was studied using this method.Methods:a) The concentration of berberine hydrochloride in the plasma was assayed by the HPLC method.Then according to the result to describe the arcuation of pharmacokinetics.b) Choose the best chromatogram condition.Made sure of the ratio and the flow rate of the mobile phase and the temperature of the colume,selected the best internal standard.c) The preparation of the plasma samples.Take 0.5ml plasma samples were separated by centrifugation at 4000 rpm for ten min.The upper clear solution was transferred to a tube entirely and dried by N2, 0.5ml ether was added to it and dried by N2 again. And then methanol was added to it and injected of each sample into the instrument.d) The selectivity of the method: Each of the berberine hydrochloride solution ,blank plasma,plasma with berberine hydrochloride,actual sample was injected to the instrument and the chromatogram of them was recorded respectively.e) Preparation of standard curve : A set of berberine hydrochloride solution was added to several blank plasma and was injected to the instrument after treated.Standard crurve was made with the content of berberine hydrochloride as abscissa and the ratio of peak area as ordinate and the regression equation was got .Make sure of the range of it.f) Precision:The 5 piece of same plasma samples was assayed unremitly to determine the...
