Purpose:To investigate the hyperglycemic effects of mangiferin combined with berberine hydrochloride(BM)on blood glucose in GK(Goto-Kakizaki) rats model. And to built a sensitive specific RP-HPLC method for the determination of mangiferin and berberine hydrochloride simultaneously in rabbit plasma.Method:1. To mix mangiferin and berberine hydrochloride at the rate of 1:1,and to observe the hyperglycemic effects on the GK rats model. 2.After mixing with the internal standard(IS) 4-(dimethylamino)benzaldehyde.The separation was carried out on a C18 column(4.6mm×250mm.5μm) using a mixture fo water (contenting o.7% phosphoric acid) and methanol(60:40,v/v/v)as mobile phase delieverd at 0.5ml·min-1.The rats were given with mangiferin or mangiferin combined with berberine hydrochloride,respectively.And the plasma samples were extracted with acetonitrile. The plasma concentration was determinated by the RP-HPLC method and the pharmacokinetic parameters were analyzed.Result: 1.The changes of rats body weight and the blood glucose were significantly decreased. 2.The pharmacokinetic process of Mangiferin single dose or combined with betberine were described as three-compartment model.The parameters were t1/2 :(22.94±6.85)h;Cmax ( 1191.85±149.84 )μg·L-1;Tmax 3h ; AUC0-t (6676.45±322.70)μg·h·L-1;AUC0-∞(20855.15±1538.83)μg·h·L-1; t1/2:(19.80±0.88)h;Cmax(727.33±45.14)μg·L-1;Tmax 3h;AUC0-t (8287.33±131.99)μg·h·L-1;AUC0-∞(25149.80±1982.61)μg·h·L-1。And the process of berberine hydrochloride was described as two-compartment model. The parameters were t1/2:(22.94±6.85)h;Cmax(1146.23±245.38)μg·L-1;Tmax (0.83±0.26)h;AUC0-t (3595.13±342.09)μg·h·L-1;AUC0-∞(7047.87±213.30)μg·h·L-1.Conclusion: The body weight of negative control group significantly decreased,and continuely droped during aderministration of water,the body weight of positive group and BM 20.0,40.0mg?kg-1 groupwere increased, and the blood glucose were significantly decreased.When the mangiferin was combined with berberine hydrochloride, the pharmacokinetcs of mangiferin was changed significantly.
