Objective: To evaluate the effects of Fogarty ballon catheter thrombectomy on rabbit venous endothelial morphology and function of the Non-thrombus vein segment and provid theoretical foundation for the therapy of deep vein thrombosis.Methods: There are 80 experimental rabbits, which had been divided into 4 groups randomly: Shame operation group(Group SG, n=25),Thrombosis group(Group TS, n=25),Thrombectomy group( Group TG, n=25)and Normal group(Group NG, n=5).The poster vena cava under renal vein of 50 experimental rabbits were ligated for 48 hours to induce thrombosis model .These thrombosed veins were medially separated to Group TS and Group TG, which group have 25 rabbits.Group TG was performed with 2-F Fogarty ballon catheter.Group SG were just opened and closed the venter as controls. Collected the sample of double cervical vein at specific points,which are 24 hours, 4days, 14days, 28days after the operation. The structural ingrity of the endothelial layer was assessed by means of scanning electron microscopy and transmission electron microscopy. The Mrna level plasminogen activated inhibitor-1(PAI-1)and tissre factor(TF) expressed by rabbit endothelial cells in four groups were analysised by fluorescence quantitative RT-PCR(FQRT-PCR).Results: Scanning electron microscopy results that the endothelium of NG group was normal, in SG group, the injury of endothelium was slim, reachedâ… on days 7; in TS group ,the injury degree of endothelial cell was aggravated gradually ,reachedⅡ~Ⅲ; in TG group, the injury degree of endothelial was aggravated first, on days 7, the injury reached the peak,that wasⅡ~Ⅲ, then the endothelial began to repair,on days... |
