| This thesis choosed horseradish peroxidase(HRP) and hemoglobin(Hb) as typical enzyme of peroxidase and mimetic peroxidase . Researched the application of two enzyme in the clinic analysis with molecular emission spectrometry (chemiluminescence and fluorescence) by flow injection analysis(FIA) . The glucose and IgG in human serum were determined and the results were satisfactory .1 , With the catalyst of hemoglobin, the fluorescence was yielded by the p-cresol and H2O2 that the glucose was catalyzed by the glucoxidase and released. Because of these, a new indirect method detecting glucose of human serum was established by using FIA. The sampling frequency was 23 samples/h. Glucose was quantified in the linear of 3.0×10-5 mol ·L-1 3.0×10-4 mol ·L-1 with the limit of detection of2, With the catalyst of hemoglobin , the chemiluminescence was yielded by the luminal and H2O2 that the glucose was catalyzed by the glucoxidase and released . Put the immunoaffinity column that fixed IgG in front of the chemiluminescence detector. In the process of preincubation , the free HRP labeled anti-IgG that not bounded with sample IgG , throughed the flow system and reached the immunoaffinity column, the free HRP labeled anti-IgG reacted with IgG that fixed in the immunoaffinity column because of the specific combination between antibody and antigen . Detected indirectly IgG in the human serum by the intensity of chemiluminescence that catalyzed by HRP labeled anti-IgG which bounded in the immunoaffinity column.
