Immunoassay with electrochemical detection is an attractive method due to its high sensitivity and low detection limit. It can also compensate its deficiency in differentiation with immunity. So it is of interest because of wide detection range and low detection limit. It is suitable for the analysis of small amounts, the instruments are simple, and the operation is convenient. So the linear sweep voltammetric was used to study the 3,4-Dianimobenzoic acid-H2O2-horseradish peroxidase (HRP) voltammetric enzyme-linked immunoassay new system. So far, the systems were not repoted by others. The optimal condition of enzyme-catalyzed reaction were investigated, and the best reaction condition of these systems were selected. In addition, the virus clinico detection was also performed. The 3,4-Dianimobenzoic acid-H2O2-horseradish peroxidase (HRP) voltammetric enzyme-linked immunoassay new system had extremely high sensitivity. HRP can be measured with a detection limit of 6.0×10-13 g/mL and a linear range of 4.0×10-9 4.0×10-12 g/mL.This new voltmmetric enzyme-linked immunossy systerm has firstly been successfully applied to the detection of αFP. The linear range is 0.20-50 ng/mL. Under the optimum conditions, the human serum samples were detected. The results showed good corresponding relationship with those in spectrophotometric ELAISA method.This new voltmmetric enzyme-linked immunossy systerm has firstly been successfully applied to the detection of CEA. The linear range is 0.50-80 ng/mL.Under the optimum conditions,the human serum samples are detected.The results have good corresponding relationship with those in spectrophotometric ELIS A method.
