Effects Of Niacin On The Expression Of CAMP And ABCA1 In PBMCs

Objective: Observing the effects of niacin on the expression of cAMP and ABCA1 in PBMCs which incubated in vitro,and investigating the correlation of niacin adjusting the expression of ABCA1 and cAMP/PKA pathway .Methods: The patients were divided into 3 groups according to the coronary artery angiography and blood lipid inspection: 1. Non-CHD patients' group (as control);2. High blood lipid CHD patients' group;3. Normal blood lipid CHD patients' group;and the 2nd and 3rd groups were divided into four sub-groups respectively: control, niacin, PKA inhibitor, niacin+PKA inhibitor .Aseptic gathering femoral artery blood, and the blood was drawn to isolate PBMCs to incubate. All the control groups do not add any intervention factors, each group of cells were incubated for 3 ~48 hours in each medium. Selects 3hr, 12hr, 24hr, 48hr four time points,using reverse transcription polymerase chain reaction and western blot protein signature examines the expression of ABCA1 mRNA and ABCA1 protein respectively, using low pH EIA examins the cAMP content in PBMCs,and investigate the effects of niacin on the expression of ABCA1 and cAMP in PBMCs.Results: â‘ Each group PBMCs'expression of ABCA1 and the cAMP level changesduring 3h~48h does not have the obvious difference (P>0.05); â‘¡ The expression of ABCA1 of CHD high blood lipid patients' in PBMCs compares the non-CHD patients' obviously to reduce (P<0.05),but in the CHD normal blood lipid patients' the expression of ABCA1 in PBMCs compares the non-CHD patients' not to have the obvious change (P>0.05); â‘¢ The niacin alone affects obviously increase the CHD high blood lipid patients' PBMCs the ABCA1 expression (P<0.05),at the same time makes the cAMP content in the PBMCs to increase (P<0.05); â‘£ PKA inhibitor Ro 31-8220 alone affects can obviously reduce CHD normal and high blood lipid patients'expression of ABCA1 in PBMCs (P<0.05),at the same time reduces the cAMP content in PBMCs (P<0.05); (5) In the niacin +PKA inhibitor group,the effects of niacin increases the...
