Objective Discuss the relationship between the obesity and the injury of vascular endothelial ; observe the changes of the serum lipids and the cardiovascular ultrastructure in rat of obesity , and research on the effect of tanshinoneâ…¡A ( Tanâ…¡A) and niacin with different dosage on the cardiovascular function of infant obese rat.Method The obesity infant rat was divided into eight groups: control group, higher fatty acid group, low Tanâ…¡A group, middle Tanâ…¡A group, high Tanâ…¡A group, low niacin group, middle niacin group, high niacin group. All rats were fed by fat-rich-diet except the normal control group. The intervention groups were treated by different dosage of corresponding drug when the animal model of obesity had been made, while the normal group was treated by saline. All rats were sacrificed after thirty days of intervention. The level of riglyceride(TG), total cholesterol(TCH), high density lipoprotein(HDL), low density lipoprotejn(LDL), very low density lipoprotein (VLDL), PGI2, TXA2 and IGF-1 factor were measured, and the tissues of aorta thoracica and myocardium were observed by transmission electron microscope (TEM).Results (1) positive effect of Tanâ…¡A and niacin on the hyperlipidemia was observed, especially on the level of HDL-C; (2) The level of IGF-1 in the serum after Tanâ…¡A treatment was slightly increased, the effect of niacin was weak. (3) positive effect of Tanâ…¡A and low niacin on the disbalance ratio between TXA2and PGI2 was observed, and it could protect vessel endothelium. (4) The injury of Tanâ…¡A on the rat myocardium was weaker than the higher fatty acid group, especially the low and middle dosage of Tsnâ…¡A. This effect of high niacin was not obvious. (5) The injury of Tanâ…¡A and niacin with low and middle dosage on the endothelium cell was weaker than the higher fatty acid group, This effect of high Tanâ…¡A was obvious, and no effect of high niacin was observed.Conclusion Protection effect on the vessel endothelium was obviously with low dosage of Tanâ…¡A and niacin, and it could meliorate the injury of myocardium by obesity. Conclusion It is obviously that enough dosage of Tanâ…¡A and low dose niacin have good effect on regulating the lipid of blood in this experiment. LDL after oxidized modification may damage the blood vessel endothelium, HDL may protect the blood vessel endothelium. These two drugs would effect on elevating the HDL-C level. Meanwhile, these may reduce the level of TG, Tch, LDL-C and the VLDL in some degree. Therefore, it has obvious regulation effect on the blood serum cholesterol in the experimental hyperlipemia rat. IGF-1 is a kind of active protein peptide material, and functions on vasorelaxation. TXA2 and PGI2 are one couple of important index on the secretion fuction of vessel endothelium. The IGF-1 expression level reduces obviously in the high fat control group comparing with that in the normal group. After Tanâ…¡A intervening for thirty days, the IGF-1 expression level in the rat blood serum has equally elevated, but the extent is small, the changes is not obviously. The effect of niacin is weak. The balance between TXA2 and PGI2 is important for maintaining the normal blood vessel function and for protecting endothelial cell. TXB2 and 6-keto-PGF 1a are the stable metabolite of TXA2 and PGI2 in the blood plasma, respectively. Therefore, to determine the TXB2 and 6-keto-PGF1a level in the blood plasma usually reflects the TXA2 and PGI2 content because the TXA2 and PGI2 are extremely unstable in the blood plasma. The TXB2 level in rat blood serum of the intervention group is lower than that in the control group, especially in the Tanâ…¡A and niacin group. The 6-Keto-PGF la level of blood serum in each intervention group is also higher than that in the control group, especially in the Tanâ…¡A group. So, Tanâ…¡A and small dosage of niacin affect the disproportionality of TXA2 to PGI2, then protect the blood vessel endothelium.In summary, low dose Tanâ…¡A and the niacin may improve the damage of cardiac muscle induced by experimental obese and protect the blood vessel endothelium.
