| Natrix tigrina Lateralis (NTL) is nontoxic snake belonging to Colubridae ,is also named neck-pheasant, it is one of the zonic medicinal materials with important medicinal value. NTL has the function of expelling wind, eliminating dampness, relieving pain due to rheumatism, detoxification, etc., and because of its remarkable curative effect to rheumatoid arthritis and bone tuberculosis, and so forth, it has been applied far and wide in Chinese traditional folk. Currenly, there is no report on the systematic study of analgesic effect and mechanism of analgesia.Our experiment is based on the study of the anti-inflammation action and mechanism of anti- inflammation that have followed to accept my tutor' s research for many years,we investigate the analgesic effect of NTL ethanol extract to central nerous or peripheral nerve in rats or in mice through using several experimental study models to quantitative pain, and its mechanism of analgesic is discussed. The purpose is to provide a scientific experimental basis for clinical development and further research. Method and results are as follows:1.The experimental study of analgesic effect to central nerousThis experiment opens out that NTL ethanol extract can inhibit the pain action of licking hind paw caused by hot plate in mice, the pain action of tail-flicking caused by heat stimulus in mice,and the pain action of pressing tail-root in mice through experimental study of effect of NTL on licking hind paw caused by hot plate in mice, tail-flicking caused by heat stimulus in mice,and pressing tail-root in mice ,it can obviously prolong the incubation period of lickinghind paw caused by hot plate and tail-flicking caused by heat stimulus in mice ,and there is significant difference between NTL and control group, in a dose-dependent manner.It was concluded that the analgesic effect of NTL might be related to the central nervous system.2.The experimental study of analgesic effect to peripheral nerveThis experiment opens out that NTL ethanol extract can inhibit the pain action of writhing induced by acetic acid in mice and the pain action of formaldehyde- induced pain in mice through experimental study of effect of NTL on writhing induced by acetic acid in mice and on formaldehyde- induced pain in mice, and there is significant difference between NTL and control group, in a dose-dependent manner.lt was concluded that the analgesic effect of NTL might be related to peripheral nerve system.3. The effect to monoamine neural transmitterIn the assay of the content about the monoamine neural transmitter—5-HT> NE> 5-HIAA in blood and in brainste of rats, which shows that NTL effects antinociceptive in central nervous through exciting the liberation of monoamine neural transmitter — 5-HT and inhibiting the liberation of monoamine neural transmitter — NE in brainste;and effects antinociceptive in peripheral nerve system through inhibiting the liberation of neural transmitter — 5-HT, NE> 5-HIAA in blood.To sum up, the experimental study on the analgesic function and its mechanism of NTL shows that it might be effect the analgesic function in peripheral nerve system like the non-steroidal anti-inflammatory drugs,and can similialy effect analgesic functionin central nervous system through inhibiting and exciting the liberation of several monoamine neural transmitter in brainste.Through the experimental study above,the analgesic characteristic and mechanism of NTL is given basically overall evaluation and explanation, and its traditional function of expelling wind, eliminating dampness, relieving pain due to rheumatism and treating rheumatism is tested elementarily, which provides stable scientific basis for reasonable exploiture and use of NTL. |
