Objective: To evaluate the prognostic significance of c-erbB-2's expression gastric carcinoma and to provide the theroretical basis for anticipating the prognosis and choose therapeutical drug.Methods:1. Literatures which published during 1990-2005 were identified from MEDLINE,PUBMED,OVID,CNKI,CBMdisc,Wanfang,Weipu Database with key words as Gastric cancer(gastric carcinoma),c-erbB-2(c-erbB2,HER-2,HER-2/neu,HER2,neu), language restricted to Chinese and English. References of selected literatures were checked at the same time. Criteria of selecting literature: gastric carcinoma were primary and the time of follow-up was more than 5 years; the articles are related to c-erbB-2 expression /gene amplication and prognosis of gastric carcinoma ; the data were sufficient. Data from literatures were arranged and analyzed ,the following data were extracted:the number of survival of c-erbB-2 positive and negative by the end of the 5 year follow-up, then the database was established.The stastistical analysis was performed by software RevMan 4.2 which was provided by the Cochrane Collaboration. Heterogeneity evaluation of all chosen literatures was done . P<0.05 was considered statistically significant. The effective models were selected. Results of analysis were expressed by odds ratio(OR) for the variable category.2. The immunohistochemical method was used to detect the expression of c-erbB-2 in gastric carcinomas which time of follow–up was more than 5 years during 1999.10-2001.5 and the clinical data were sufficient. Survival was supposed as the prognostic index. The relation between the prognosic significance of c-erbB-2 and the clinicopathological parameters in gastric carcinoma was analyzed.3. The results of the second part of our study and the 11 chosen literatures were analyzed by meta-analysis.Results:1. 76 literatures were searched from MEDLINE,PUBMED,OVID,CNKI,CBMdisc,Wanfang,Weipu Database.11 of 76 literatures matched our selecting criteria.The heterogeneity evaluation of 11 chosen literatures showed no significant (P=0.12). So the fixed effect model was used to analyze the homogeneous literatures. The pooled odds ratio(OR) of 5-year survival rates of 11 literatures was 0.69(95%CI:0.54-0.87).The pooled odds ratio(OR) of three literatures from eleven literatures was 0.67(95%CI:0.48-0.94). 5-year survival rates...
