[Objectives]Screen serum protein expression maps in patients with non-Hodgkin lymphoma (NHL) by using surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS), and to find the biomarkers in NHL for early stage diagnosis.[Methods] Using immunohistochemical method to investigate the expression of ALK1, bcl-6, CD3, CD5, CD15, CD20, CD23, CD30, CD56, CD79α, PAX5 and TdT in the lymphoma tissues of 40 NHL, which were classified according to WHO classification Tumours of Haematopoietic and Lymphoid Tissues, and using SELDI-TOF-MS to analyze sera of these cases on WCX2 protein chip. The data was analyzed by Biomarker Wizard software.[Results] 40 cases was diagnosised and classified specifically according the immunohis- tochemical method, including 27 B-cell lymphoma and 13 T–cell lymphoma. 157 protein peaks were detected in the molecular range of 2-20k, among which 5 ones were significantly different between T and B-cell lymphoma(p<0.05). Peaks of 8083,15944,7979,16110 and 8057 Da were upregulated in B-cell lymphoma cases.[Conclusions]The specific early stage biomarkers of NHL could be detected using immunohis- tochemical method and SELDI-TOF-MS, which had a bright prospect of clinical application.
