Purposes: (1) To find the most suitable protein concentrations. (2) To discover the proteins with relation to TP and provide new ideas of finding new biomakers of TP;(3) To establish initially diagnostic models of TP.Methods: (1) To collect Pleural effusion samples of 22 cases of TP, 16 cases of LCPM , measure all the pleural effusion samples for protein concentrations by means of Biuret method , find the highest concentration of all the samples which is 70.0μg/μl, and regulate the concentration to different ones by adding different volume buffer,then to add the sample onto the protein chip for each protein concentration (2) To treat 38 samples according the most suitable protein concentrations,then to add the sample onto the protein chip for each case (3)Special protein or peptide spectra was determined by SELDI-TOF-MS measurement . (4) To compare the protein profiling results of different protein concentrations.(5)Ciphergen Biosystems software was used to analyze the proteomic profiles in order to find proteins relating to tuberculous pleurisy and establish initially the diagnostic pattern.Results: (1)Analysis of pleural effusion protein fingerprints can conclude more significant difference protein peaks was detected when the concentration is 600μg/390μl.(2)There were 147 protein peaks at the key M/Z values range from 2000 to 8000 Da. Among them 14 protein peaks show the difference obviously by the software analysis(P<0.05).
