Objective:To study the expressions of nm23, P53 in gastric cancer cell line SGC-7901 with Arsenic Trioxide(As2O3) , 5-Fu and combinating the drugs.Methods: Human gastric cancer cell line SGC-7901 was serial-subcultivated in the RPMI1640 culture fluid including 10%FCS , 100U/ml penicillin and 100U/ml streptomycin in the incubator demanding 37℃, 5% CO2 and saturated humidity;The log phase cell line was induced with As2IO3 and 5-Fu, besides to set up blank contral group, As2O3-group and 5-Fu-group;After defferent period, to detect the effects of growth in the cell lines by MTT Method;To detect the expressions of nm23, P53 in the cell lines by P-V method;To analyze the results with VIDAS image analysator according to absolute gray scales.Results: 1,The growth of the three non-blank groups was inhibited remarkably,and the effects of inhibition in the three groups present time-effect relationships;The effect of inhibition in the combining- drug group was more powerful than As2O3-group and 5-Fu -group;The effects of inhibition in the As2O3-rgroup and 5-Fu-group was similar. 2, Differences of the expressions of nm23 between 4 groups had notable statistical significance(P<0.01), the expression of nm23 in blank contral group was lowest;In the three non-blank groups, the expressions of nm23 was higher notably than in blank contral group(P<0.05),but the differences of the expressions in the three non-blank groups had no statistical significance(P>0.05).3, Differences of the expressions of P53 between 4 groups had notable statistical significance(P<0.01), the expression of P53 in blank contral group was highest;In the three non-blank groups, the expressions of P53 was lower notably than in blank contral group(P<0.05),but the differences of the expressions in the three non-blank groups had no statistical significance(P>0.05).Conclusion: 1, As2O3 and 5-Fu can both inhibit the growth of the cell line,the effects of inhibition is similar, but synergistic use of As2O3 and 5-Fu can improve the efficacy .2^ As2O:i can up-regulate the expression of nm23,and down-regulate the expression of P53.
