Objestive ①To investigate the effect of hyperthermic chemotherapy on apoptosis and its mechanism of liver cancer cells BEL-7402. ② To investigate the effects of5-fluorourail and mitomycin c combined with 42℃ whole-body hyperthermia for advanced hepatic carcinoma.Methods ①Human liver cancer cell lines BEL-7402 were cultured in vitro andunderwent hyperthermic chemotherapy in 42℃. The cell growth and apoptosis were observed,the cell cycle were studied by flow cytometry,and the expression level of apoptosis regulator gene were examined by immunohistochemical method.Results②16 patients with advanced hepatic carcinoma enrolled in the trial group,5-Fluorourailand mitomycin c were infused during the treatment of whole-body hyperthermia. 26 patients in control group received chemotherapy only.Results ①The effect of hyperthermic chemotherapy on liver cancer cells BEL-7402 was remarkable.The G1 phase arrest and the upregulated of bax and Fas were observed after hyperthermic chemotherapy.② The response rate (WHO standard) were 31.25% and 3.85% respectively.Conclusions ①The apoptosis rate induced by hyperthermic chemotherapy is higher than that of chemotherapy.The cell cycle arrest and expression of apoptosis regulatior gene may contribute to the higher level of apoptosis. ② This clinical trialdemonstrates the feasibility of performing whole-body hyperthermia combined with chemotherapy.
