Objectives : This research work was to investigate enzyme activity of Paraoxonase-l(PON1) and genotype of PON1A/B192 among thrombotic cerebral infarction individuals under 45years old. The genotype and enzyme activity would be determined if they were associated with the disease among individuals under 45years old. This work would discuss the significance of PON1A/B192 genotype and enzyme activity, moreover, provide some theoretical evidences.Methods: Case-control study was adopted. Phenol acetate was used as substrate to determine the enzyme activity of PON1. Genotype of PON1A/B192 was defined by PCR-RFLP. Thrombotic cerebral infarction related causes/clues were detected via biochemistry methods (e.g. TG, TC, HDL-C, apoA, apoB, Lp(a)) and questionnaires.Results:1.Of routine hazardous factors e.g. blood pressure, high pressure history, diabetes history, familial thrombotic cerebral infarction and smoking history, patients show higher then control group with P<0.05 or more. Plasma lipid index of the two group showed difference of different extent with TG, TC, HDL-C the most difference (P<0.01).2 Genotype of each individual in the two groups was determined by PCR-RFLP and gene frequency was calculated. A/B gene frequency of control group were 42%/58% separately while the data in patient group were 30%/70% (P<0.05). Frequency of BB genotype in patient group was higher than control group (P<0.05). PON1 enzyme activity was determined case by case, which showed difference in patient group and control group with P<0.01 while BB genotype of patient group showed lower enzyme activity than AA and AB genotype.3.Plasma lipid index, BS, BMI and blood pressure was contrasted among different PON1A/B192 genotypes with no difference. Factors with obvious difference in single factor analysis (smoking, high pressure, TG, TC, PON1 enzyme activity, BB genotype, HDL-C, familial history, ApoAl, ApoB) were draw into logistic regression, and only genotype BB, PON1 enzyme activity, high pressure, high plasma lipid, diabetes history, smoking and familial thrombotic cerebral infarction entered the logistic model, which indicate that they are hazardous factors of thrombotic cerebral infarction under 45years old.ConclusionsrOther than well-known hazardous factors e.g. TG, TC, HDL-C, high pressure, diabetes, smoking, familial history, PON 1 A/B 192 genotype was associated with thrombotic cerebral infarction under 45years old. This research work alsoindicate that P0N1 enzyme activity should be an important biochemistry hazardous factor in evaluating and inspecting young thrombotic cerebral infarction patients. Meanwhile, PON1A/B192 genotype was an important clue/cause in young thrombotic cerebral infarction patients. We hold that inspection of the two index would prevent young thrombotic cerebral infarction to some extent.
