Objective: The chemical constituents of the ethanol extract of Rubia yunnanensis Diels. were studied, and through this study , the optimum extract condition for Rubia yunnanensis Diels. was established.Materials and methods: 1. Rubia yunnanensis Diels. was extracted with ethanol and isolated with silica gel column chromatography. The constituents were identified on the basis of spectral data. 2.The sum content of anthraquinone determined by spectrophotometer and the weight of the crude extract were used as the result indexes of the orthogonal design L9 (34) to establish the optimum extract condition for Rubia yunnanensis Diels.Results: 1. Three known constituents were isolated and their structures were identified as 1.3.6-trihydroxy-2-methyl-anthraquinone; Rubiarbonot A; β-sitosterol. 2 .The optimum extracting condition of Rubia yunnanensis Diets, is: 8 times amount of 70% ethanol, and refluxing at the temperature of 80 ℃ for 90 min twice.Conclusions: The chemical constituents of Rubia yunnanensis Diels. are complicate, it's hard to establish the optimum extract condition by using a single compound as the result index. The active ingredients are maximally extracted with good reproducibility by using the sum content of anthraquinone and the weight of the crude extract as the result indexes of the extract.
