| Objective and background—dilated cardiomyopathy(DCM) is a common form of heart disease, mutations of genes encoding myocardium and other proteins have been identified in a part of patients with idiopathic DCM. A recent study confirmed that mutations of the metavinculin-specific exon 19 of vinculin gene caused IDCM. Our test was to screen whether the mutations of the metavinculin-specific exon 19 of vinculin gene was one of the causes of Chinese idiopathic DCM population.Methods and Results—110 DCM patients and 110 control cases were included in our study. The metavinculin-specific exon 19 of vinculin gene was analysed by polymerase chain reaction(PCR), polyacrylamide gel electrophoresis(PAGE), degenerated high performance liquid chromatogram(DHPLC) and DNA sequence analysis. No mutations related of the metavinculin-specific exon 19 of vinculin gene had been identified in all of the subjects, except of a common, previously reported 2814C—-G neutral polymorphism(Gly938Gly) in one patient.Conclusions—In Chinese idiopathic DCM population, no mutation of the metavinculin-specific exon 19 of Vinculin gene was found. |
