A Study Of HMG-CoA Reductase Inhibitors Effect On Endothelium Function In C57BL/6 Mice

Objective: We build the model of carotid stenosis on C57BL/6 mice through place collars on one side around the common carotid artery. Then give high-cholesterol diet 12 weeks to see if the stenosis could make atherosclerosis (AS) formation. And as well as we give atorvastatin and/or fosinopril through drinking water to find if the medicine could do some effect on endothelium function, carotid atherosclerosis formation and development.Method: 40 female C57BL/6 mice were 7 to 8 weeks old at the time of entry the study. Divided into four groups. Both carotid sheaths were opened and the common carotid arteries were dissected free from the surrounding connective tissue, avoiding damage to the vagus nerves and the carotid bodies. In the left side, a 5mm long and 0.3mm inside diameter collars were placed around the common carotid arteries, and their axial edges were approximated by placement of 3 circumferential silk ties, and the right was the sham-operated side. From the beginning, each group was given high-cholesterol diet, and three of them were divided given atorvastatin or fosinopril or atorvastatin and fosinopril. After 12 weeks, take both sides of carotid artery and use hematoxylin and eosin-stained sections for morphometric analysis. Measure and compare the serum level of cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), nitric oxide synthetase (NOS), endothelium nitric oxide synthetase (eNOS), inducible nitric oxide synthetase (iNOS), superoxide dismutase (SOD), maleic dialdehyde (MDA), macrophage colony stimulating factor (M-CSF).Result: 1 , Through the HE dyeing we see the early atherosclerosis formation on the left side of the contrast group which express as the intima thickness, intima-media ratios(IMR) increase, lumen diameter decrease, inflammation cell were found under the endothelium et al. But no atherosclerosis was found in the common carotid arteries of sham-operated sides and the both sides of the other three groups.2 , After 12 weeks we found the serum level TC and LDL-C increase...
