Objective: The aim of study was to explore the relation between LEP level in blood serum,LEP receptor variation ,blood lipid,diet,breeding, etc and breast cancer, and to find out the risk factors of beast cancer Methods: A case-control study was used. There was 94 primary female breast cancer patients who were cured in tumor hospital of Shanxi from 2004-07 to 2004-12 in case, and 128 healthy people came from countryside,county and town in control. All objects were investigated including height,weight,waistline,breechline and blood pressure; 2ml anticoagulant blood and 2ml non-anticoagulant blood from elbow vein of investigation object were collected. The DNA was extracted from anticoagulant blood, the polymorphism of Gln223Arg was detected by PCR-RFLP;blood serum was isolated from non-anticoagulant blood, and assayed the level of blood sugar and blood lipid by Automatic biochemistry analyzer. Results: â‘ Multiple Logistic regression analysis for doubtful risk factors: Agents were introducted, which came from mono-factor analysis and literature. The agent that can enter multiple Logistic regression analysis as follows: LEP leve(lOR=1.53),LEP receptor variation(OR=4.87),frequency of fruit edible(OR=0.58),frequency of vegetable edible(OR=2.06),quantity of flour edible(OR=0.64),quantity of shrimp edible(OR=0.31),well history of mammary gland disease(OR=3.15),history of mammary gland trauma(OR=9.85),law of menstruation(OR=3.03),history of contraceptive take(OR=6.44),free cholesterol of blood serum(OR=15.19),Triglyceride of blood serum(OR=5.54),Apolipoprotein A1 of blood serum(OR=12.73),Apolipoprotein B of blood serum ( OR=0.05 ) and waist-to-hip ratio(OR=3.68). â‘¡Relationship of agents:There was positive correlation between LEP level and waist-to-hip ratio and body lipid exponent, its correlation coefficient was 0.423,0.468 respectively. LEP level of different average income was significant statistic difference(P<0.001). The blood lipid level of high income was higher than the low incomes, but only the difference of Apolipoprotein A1 was significantly(P=0.025).The frequency of vegetable edible of high earnings was higher than the low earnings(P=0.038),and trend test was statistic significance(P<0.05). the quantity of flour edible of high income was less than low incomes(P=0.046),and trend test was statistic significance(P<0.05). â‘¢Interaction analysis:There was interaction between LEP level and free cholesterol of blood serum(S=2.52), API=0.44;There was interaction between LEP level and Apolipoprotein A1 of blood serum(S=1.83), API=0.41. Conclusion: 1) LEP level was independent risk factor of occurrence of breast cancer, so LEP receptor variation did. 2) The free cholesterol of blood serum,Triglyceride and Apolipoprotein A1 were risk factors of occurrence of breast cancer. 3) There was interaction between LEP level and free cholesterol of blood serum,Apolipoprotein A1. 4) Maladjustment of menstruation,usage of estrin,may be risk factors of occurrence of breast cancer,well history of mammary gland disease and history of mammary gland trauma may be the risk factors of occurrence of breast cancer. The shrimp and fruit were protective factors ofoccurrence of breast cancer. 5) The influence of vegetable and flour to breast cancer was due to the difference income.
