Objectives After feeding the high fatty diet SD rats with Anemarrhena asphodeloides Bge, assaying the blood fat with kit and the LDLR level of their livers by immunohistochemical method, then analyze the Effects of sapogenins in Anemarrhena asphodeloides Bge on the blood fat of hyperlipemia rats and the activity of the low-density lipoprotein receptor of the liver cell membrane and discuss the mechanism of regulating the blood fat.Methods Divide 80 male SD albino rats into 4 groups at random, with NO. 1 called normal control group, NO.2 called model group, NO.3 called Simvastatin group and NO.4 called sapogenins in Anemarrhena asphodeloides Bge group. Feed Group 1,2 with normal animal feeds, others with high fat feeds. Feed Groupl with normal sodium (NS), Group3 with dose of 12mg/kg, Group4 with dose of 50mg/kg. 50days later, draw blood from the ventricles and assay the blood fat, then analyze the data by variance analysis through SPSSIO.O software. Make the right lobe of the liver into frozen section by OCT embedding. Detect the LDLR expression of the liver frozen section by immunohistochemical method and observe it under microscope, then to make semiquantitative analysis by computer image analysis system. The data should be expressed by means ± standard deviation and compare the groups by analysis of variance analysis.Results Compare with Group2,TC and LDL-C reduce greatly in Group4(TC and LDL-C, p <0.001).Them is not much difference between Group3 and Group4(p>0.05).The gray scale of the brown particle diameter of the liver section in Group4 exceed Group2(p<0.001),while compared with Group3,there is not much difference(p>0.05).Conclusions sapogenins in Anemarrhena asphodeloides Bge possibly enhance the expression of LDLR gene in liver, promote the synthesis of the LDLR protein and raise the quantity and activity of the LDLR and accordingly enhance the metabolism of blood fat, thus regulate the blood fat level.
