| Purpose:To study the freduency of CCR5 A32 mutation and the expression of human herpesvirus-8 latent -associated nuclear antibody in Kaposi' sarcoma patients ,investigate the pothogenesis of HHV8 in the development of XinJiang KS. Methods:Immunohistochemistory was employed to detect the LANA in Xinjiang KS of 43 cases of biopsy with paraffin embeded. HHV8 DNA and CCR5 gene mutation was detected by PCR in 68 cases of KS and in 45 cases of normal control group. Results:1)25 cases of 43 KS tissues showed strong,diffuse ,nuclear staining in spindle cells(58%),nodular and plaque lesions generally had more positive spindle cells compared to patch lesions; 15 cases of patch lesions with nonspecific inflammatory reaction, 1 case of patch lesions with obvious dermal edema and 2 cases of nodular with predominant inflammatory reaction were negitive for LANA. None of control group was positive, in statistical analysis ,the difference was significant (x~2=16.9, p<0.01)2) In 65 cases of KS, 58 had the proportions wt/wt genotypes, 7 with wt/A32 genotypes and no one with A32/A32; The proportions of wt/wt , wt/A32 genotypes were 89.23%, 10.76% respectively, compared with control group, the discrepancy of constituent ratio was not significant(x~2=1.75, p>0.05).3) HHV8 DNA were detected in 60 of 65 cases with KS, 11/45 in control group.4) In HHV8 positive group , the proportion of wt/wt and wt/A32 genotypes were 91.55%, 8.45%, respectively ,compared with HHV8 negative group, there was not significant differece(x~2=0.41,p>0.05).5) In 110 cases including KS and control group, The frequency of CCR5 A32 mutant alleles was 3.64%.conclusion:1) XinJiang KS has special clinicpathological characteristics compared to the general features of KS.2) LANA may play a important role in the nodular and plaques stage of KS; the negative expression of LANA in inflammatory ks tissues indicated the change of immune state may affect the expression of virus gene.3) The immunohistochemical detection of LANA in the nodular and plaques stage of KS is a sensitivity and specificity means of differentiating KS from other vascular and nonvascular spindle cell... |
