Objective To explore the expression of anti-oncogene P53, E-cadherin and CD34 in the hepatocellular carcinoma of 41 cases, and the associaton of them with clinical pathology and the prognosis. Methods Forty-one specimens of HCCs, who were followed up, were obtained from the operative specimens of the affiliated hospital of Qing Dao University Medical College with total clinical datas. The P53,E-cadherin and CD34 were measured by the immunohistochemistry two-step methods of PV-6000. Results The five years survival rate of overall patients is 44.4%, media survival time is 42.34 months. The five years survival of small HCC (diameter≤5cm )is 66.9%, and big HCC(diameter > 5cm) is 28.2 % . The five years survival of positive groups of P53,E-cadherin,CD34 are 36 % ,44.86 % and 44.4 % respectively. The positive rate of P53,E-cadherin,CD34 in forty-one specimens of HCCs were 63.4%,48.78% and 100%respectively; the expression of P53 with AFP level and Edmondson classification both have significant correlation(P<0.05), the expression of E-cadherin decreases significantly in patients with big HCC, invasion and Edmondson classification (Ⅲ and Ⅳ) (P<0.05), the expression of CD34 with age and invasion of tumor both have significant negative-correlation(P<0.05); the survival rate in patients with negative-expression of E-cadherin decreases significantly; the expressions of P53,E-cadherin and CD34 have no significant correlation by Spearman relative analysis(P>0.05); the post-operative survival rate of the HCC patients with the positive-expression of E-cadherin and microvessel density(MVD≤40) of CD34 is more than the group of the expression with single factor and the group of non-expression, although the difference between the two groups don' t have significance (P=0.2884), but the result may relate to our few specimens, the comparison need more specimens. In both monovariate Kaplan-Meier Plot and multivariate Cox regression analysis, only the expression of E-cadherin protein is linked to unfavorable prognosis. Conclusion 1.The gene of P53 has high-expression in the tissue of HCC, and the expression changes with the different phases of HCC. 2. The expression of E-cadherin decreases significantly in patients with big HCC, invasion and Edmondson classification (HI and IV), the survival rate of negative-group decreases significant. 3. The expression of CD34 has no significant relationship with the prognosis of HCC patients, so it can' t be used for the index of prognosis. 4. The expression of E-cadherin and CD34 may relate to the prognosis of HCC patients.
