Influenza is a kind of acute respiratory track infectious disease caused by influenza virus, it threats mankind , poultry and mammal's health seriously. At present ,there is no effective therapy to the influenza and its complications.Traditional Chinese medicine (TCM) has its superiority in preventing and treating influenza . This thesis studied the therapeutical effect of Jiawei Xuanfei Toujie Ji (traditional Chinese medicine prescription)on influenza viral pneumonia in mice and some immunological mechanisms. Objective: To study the therapeutical effect of Jiawei Xuanfei Toujie Ji (JXT) on influenza viral pneumonia in mice and some immunological mechanisms. Methods: 1.ICR mice were inoculated intranasally with mouse lung-adapted Influenza virus strain A/FM/1/34(H1N1) (FM1) 25LD50, 25μl and were treated with JXT, Ribavirin was used as postive control drug. The effect of JXT on the weight ,survival rate of FM1 infected mice after 14days inoculation was observed. 2. BALB/c mice were inoculated intranasally with FM115LD50, and were treated with JXT.,at 5th day after inoculation were sacrificed ,(1) lungs were removed and weighed ,then lung index was analysised ; the virus hemagglutination tite in lungs homogenate was detected; lungs were fixed in formalin, routinely processed, and embedded in paraffin. Routine hematoxylin-and -eosin (H&E)-stained sections were examined. (2) MTT method was used to analysis T lymphocyte in mice spleens proliferation; bioactivity method was used to. detect IL-2 and IFN-γ sereted by T lymphocyte. (3)SABC immunohistochemistrial method was used to detect MCP-1,TNF-α,IL-β,IFN-γ,IL-10 expression in lung tissue sections (4) IL-2,TNF-α,IL-6and IFN-γ in mice sera were measured with ELISA( Enzyme-linked immunoadsordent assay ). Results: 1.Model group showed lower survival rate(9.66%).Compared with model group, JXT group showed higher survival rate (68.85% and 52.3%)and body weight , lower lung index and virus hemagglutination tite in lungs.2.The pathomorphology studies showed that model group lung appearance changes were consolidation and hemorrhage; the pathological changes were serious interstitial pneumonia changes, showing bronchial epithelium shedding,alveolar interval thickening,many mononuclearcells infiltrating the alveolar wall and bronchiolar wall, producing consolidated regions, serous effusion in alveolar cavities, compensatory pulmonary edema.Compared with Model group, the pathological changes of JXT group were ameliorated obviously.3. Compared with model group, the ability of T-lymphocyte proliferation and secreting IL-2 , IFN-γ of JXT group were improved obviously. 4. Model group showed higher expression of MCP-1,TNF-α,IL-β,IFN-γ,IL-10 in lungs Compared with model group , MCP-1,TNF-α,IL-β of JXT group were dramatically decreased(P<0.05),while IFN-γ,IL-10 were significantly increased (P<0.05). 4.The levels of TNF-α,IL-6 , IFN-γ and IL-4 in sera of model group were more than nomal group(P<0.01),while the level of IL-2 was lower(P<0.05) .The level of IL-10 was no obviously difference between model group and nomal group . Compared with model group, the levels of TNF-α and IL-6 of JXT were obviously decreased and the levels of IL-2,IL-10,IFN-γ were significantly elevated. Conclusions: Jiawei Xuanfei Toujie Ji(JXT) can enhance survival rate, increase body weight and reduce inflammatory pathology in the lung of FM1 infected mice.These indicate that JXT is effective for the influenza viral pneumonia. Increasing immunological fuction and regulating the productions of TNF-α, IL-6, MCP-1, IFN-γ,IL-2 and IL-10 were possible immunology mechanisms of JXT 's therapeutical effect .Regulative effects on cytokines may be one of the factors that JXT decreases the pathogenesis of influenza-induced acute lung injury.
