Background and object: Recent research shows that central nervous system can regenerate itself following injury which is the basis of functional reorganization. The new growing process of axon or dendrite is called sprouting, it is a response in which the axons of uninjured neuron go toward the injured area to replace the degenerated one. We investigated the effect of different electrical stimulation on growth associated protein-43(sprouting) and basic fibroblast growth factor and the influence of exogenous bFGF and bilateral electric stimulation on nerve regrowth in order to confirm the effect of rehabilitative treatment on regeneration of central nervous system and offered the theory basis for the clinical practice of electric stimulation and bFGF.Method:(1) Model making and group dividing:128 adult male rats (250-300g) received right middle cerebral artery occlusion, the success of model is determined by 5 degree. The 2 and 3 degree was employed in all experiments, they were divided randomly into controlled group, ipsilateral simulation group, bilateral stimulation group and bilateral stimulation +bFGF group, 32 rats in each. (2) Electric stimulation and injection of bFGF into caudal vein: The different stimulation was performed in the forelimb at the level of wrist of the subjects with 6805-C electrically heated distilling apparatus (4-5V,0.5ms,3Hz,30min per day) poststroke. Above bilateral stimulation and injection of bFGF into caudal vein within 30 min following experimental stroke were performed for the last group. (3) Behavioral testing and immunohistochemistry: 8 rats were take from each group randomly for beam walking test which shows the functional recovery, at 3,7,14,28 day, then the brain were perfused, removed and fixed. The level of optic chiasma was selected for paraffin sections of 6um, Then the sections of the brains were processed by immunohistochemistry with antibodies against GAP-43,bFGF, Then the effect of different electric stimulation and electric stimulation+bFGF on expression of GAP-43 and bFGF poststroke were monitered. (4) Statistical analysis:Statistical comparisons were performed by using one-way analysis of variance with post hoc Student-Newman-Keuls test. P values of less than 0.05 were considered significant.Results: (1) The effect of different electric stimulation on functional recovery of movement:Beam walking test shew that: electric stimulation group was different from controlled one, but the diference between ipsilateral and bilateral group is small at 3th and 7th day. At 7th day bilateral stimulation +bFGF group was statistically different from controlled and ipsilateral group(P<0.05), bilateral stimulation group and bilateral stimulation +bFGF group was increased more remarktably than ipsilateral stimulation group at 14th and 28th day(P<0.05). (2) The effect of different electric stimulation on expresstion of GAP-43: Positive product of GAP-43 exsisted in peti-infarction area, it increased at 3th day, attained the peak at 7th day, and decreased in the later days, it remained expressing at 28th day. Ipsilateral stimulation group was statistically different from controlled group at 14th day(P<0.05),bilateral stimulation group was increased more remarktably than controlled group at 7th, 14th and 28th day(P<0.05), and at 28th day different from ipsilateral stimulation group(P<0.05), bilateral stimulation +bFGF group was different from ipsilateral stimulation group statistically (P<0.05)at 3th day . (3) The effect of different electric stimulation on expresstion of bFGF: Positive product of bFGF exsisted in peti-infarction area, it increased at 3th days, and decreased in the later days, it remained expressing at 14th ,28th days.
