Expression of focal adhesion kinase(FAK) and integrin subunits a v and P 3 in colorectal carcinomaTo rearch the correlation between expression of FAK and invasion/metastasis in colorectal carcinoma(CRC), and the mechanism of interaction between FAK and integrin a v p 3 in angiogenesis of CRC,irnrnunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR) were used to detect FAK expression in 30 panels of samples including cancerous tissues, tissues surrounding tumor, 2cm(P2), 5cm(P5) far from primary tumor margin and tissues of cutting side(N). In addition, integrin subunits a v and P 3 mRNA expressions were studied on different sites mentioned above in CRC. All the results were statistically analyzed. Results show that:1. FAK expressions in tumor on mRNA and protein levels are both significantly higher than that in tissues surrounding tumor(P2N P5) and tissues of cutting side,however there is no significantly difference between tissues surrounding tumor and cutting side.2. There is significantly positive correlation between FAK expression in tumor and lymph node metastasis,neoplasia invasion, as well as Dukes' stage.3. There is no correlation between FAK mRNA and protein expressions and colorectal liver metastasis, histological type, as well as differetiation in CRC.4. Integrin subunits a v and (3 3 mRNA expressions in tumor are both significantly higher than that in tissues surrounding tumor(P2 , P5) and tissues of cutting side, however there is no significantly difference between tissues surrounding tumor and cutting side.Conclusions:1. Overexpression of FAK in tumor indicates that FAK could activate several signal transduction pathway to modulate the malignant biological behavior of CRC, including malignant transformation and tumor angiogenesis.2. The significantly positive correlation between FAK overexpression in tumor and lymph node metastasis, neoplasia invasion,as well as Dukes' stage indicates that FAK plays a key role in invasion/metastasis of CRC.3. FAK is considered the central molecule in integrin-mediated signal transduction pathway, which is closely relative to the acquirement of potential ability to invasion/metastasis of CRC. Therefore, FAK could be a prognostic predictor in CRC patients.4. Overexpression of integrin subunits a v and 3 3 mRNA could indicate that activition of the integrin-mediated signal transduction pathway via FAK contributes to angiogenesis in CRC.
