Methods: First we adopted the Kun ming pedigree mice inoculated with H22 liver cancer cells in axilla subcutaneous tissue as the transplanting model to study the effect of tumor-suppressing. Second the different doses of the alcoholic extract of Donglingcao were given to the mice orally. Finally we not only observed the changes of common status of the mice and morphologic changes of histocyte but also measured the gene expression of mutant p53, bcl-2 and bax with immunohistochemical method and the expression of activity of telomerase reverse transcriptase with hybidization in situ. Results: The results indicated that the effect of the treated groups was better than that of the control group. The study of its mechanism showed that the alcoholic extract of Donglingcao could increase the expression of bax gene and decrease the expression of bcl-2 gene, mutant p53 gene and the activity of telomerase reverse transcriptase. Conclusion: From the whole experiment, we draw our conclusion that the alcoholic extraction of Donglingcao can effectively suppress the growth of tumors. The mechanism might be related to the up-regulation of the bax gene expression and down-regulation of the bcl-2 gene ,mutant p53 gene and the activity of telomerase reverse transcriptase expression. | | Keywords/Search Tags: | Donglingcao, H22 liver cancer cell, p53, bcl-2, bax, telomerase | PDF Full Text Request | Related items |
