Object This study was to investigate the variation of plasma PAF, TXB2 and 6-K-PGF1 levels and their clinical implication in asthmatic children.Method A biochemical assay for PAF in plasma was established to determinate the plasma PAF of 30 asthmatic children at acute stage and of 24 of them at convalescence stage after treatment. Similarly, TXB2 and 6-K-PGF1 were also measured by Radioimmunoassay . The control group was 20 normal children.Result In comparison with normal group, the acute stage group showed a significant increase in PAF(155.81 62.49 vs. 56.63 40.68ug/L, P<0.01), TXB2(261. 28 90.91 vs. 1 26.09 34.54ng/L, P<0.01) and the Ratio of TXB2 /6-K-PGF1(16.25 6.01 vs. 5.26 1.24, P<0.01); the acute stage group, relative to the convalescence stage group, resulted in a statistical increase in PAF(155.81 62.49 vs. 67.36 45.57ug/L, P<0.05), TXB2(26 1.28 90.91 vs. 138.45 35.63ng/L, P<0.05) and the Ratio of TXB2 /6-K-PGFla(l 6.25 6.01 vs. 5.42 1.45, P<0.01); no significant differences were observed in the PAF(P>0.05), TXB2(P>0.05) and the Ratio of TXB2 /6-K-PGF1(P>0.05) between the convalescence stage group and the control group. There were not any significant variations(P>0.05) in plasma 6-K-PGF1 among the acute stage group (17.16 5.32ng/L), convalescence stage group(15.68 5.06ng/L) and the control normal(20.36 5.61ng/L).Conclusion Plasma PAF and TXB2 showed significantly higher atacute stage of asthma and recovered to normal levels at convalescence stage. They may be potent mediators of asthma and play important roles in immunological mechanisms of asthma.
