Objective To observe the changes of platelet activating factor and its receptor expression at the different phase of deep second-degree burn in the skins of rats. And discuss PAF and PAFR in the role of injury of deep second-degree burn wounds.Methods Take54SD rats (male) were randomly divided into control group and burn group namely;6SD rats were belonged to the control group, and burn group included48SD rats, then were randomly dvided into6groups (8/group).Take skin samples from deep second-degree burn wounds of rats at3h,6h,12h,24h,48h and72h post burns, and skin samples of the control group.The content of platelet activating factor was detected by ELISA and the expression of platelet activating factor receptor was examined by immunohistochemical techniques in these skin samples of rats, respectively. And the determination of immunohisto-chemical positive results according to semi-quantitative grading score. All experiment results carried out statistical analysis by SPSS19.0statistical software, and the measurement data were indicated with x±S; and the comparison among groups by one-factor analysis of variance; correlation coefficient t test for correlation between the two-variable.A p level less than0.05was considered statistically significant.Results (1) The level of platelet-activating factor is low in the skin tissue of normal rat, the level of platelet-activating factor increased obviously at the stage of three hours post burn and reached the peak at12hours(P<0.05), then showed downward trend from at the stage of24hours, the level of platelet-activating factor of wound is still higher than the normal skin of rat until at the stage of72hours.(2)The platelet activating factor receptor of the normal skin of rat showed negative or weakly positive expression, and its positive expression increased obviously at3hours post-burn, and the integral value of positive expression reached the peak at12hours(P<0.05), subsequently, the positive expression of PAFR weakened at24hours, but it still higher than the normal skin of rat at72hours post-burn. PAFR positive expression located in the epithelial cells membrane of sweat glands and vascular endothelial cell membrane in the skin of rat;(3) The variables of platelet-activating factor and platelet-activating factor receptor were positively correlated,and the correlation coefficient r=0.85。Conclusion The levels of platelet activating factor and the expression of platelet-activating factor receptor increased in the skins tissue of rats post burn,suggesting that platelet-activating factor and its receptor may be involved in the pathological damage process of deep second-degree burn. during the...
