OBJECTIVE We investigated the relationship between apoptosis and human uterine leiomyoma ,and the effect of estradiol and progesterone on human uterine leiomyoma apoptosis by detecting the expression of apoptopic genes Bcl-2,Bax,Fas,Fas-L in human uterine leiomyoma and in homologous normal uterine smooth muscle,and by detecting the levels of estradiol and progesterone of patients with uterine leiomyoma and women with normal menstruation.METHODS By radio-immunity analysis(RIA) we detected the levels of estradiol and progesterone of 22 proliferative phase patients with uterine leiomyoma and 30 proliferative phase normal menstrual women.By immunohistochemistry we detected the expressions of apoptopic genes Bcl-2,Bax,Fas,Fas-L in the 40 patients with uterine leiomyoma(leiomyoma group) and adjacent normal myometrium(uterine smooth muscle group).In leiomyoma group there were 22 proliferative phase and 18 secretory phase patients.RESULTS â‘ The levels of estradiol and progesterone of proliferative phase patients with uterine leiomyoma were significantly higher than those of proliferative phase normal menstrual women( ï¼°<0.01).â‘¡The expression of Bcl-2 in leiomyoma group was significantly higher than that in uterine smooth muscle group( ï¼°<0.01). The expressions of Bax,Fas,Fas-L in leiomyoma group were significantly lower than those in uterine smooth muscle group( ï¼°<0.01).â‘¢In leiomyoma group the expressions of Bcl-2,Bax,Fas,Fas-L in the proliferative phase were no significantly differences than those in the secrectory phase(P>0.05).In uterine smooth muscle group were the same(P>0.05).CONCLUSION â‘ The estradiol and progesterone were important factors to accelerate the development of human uterine leiomyoma.â‘¡It resulted in local uterine smooth muscle abnormal apoptosis that the reinforcement of the expression of the inhibiting apoptosis gene Bcl-2 and the weakeness of the expressions of the apoptopic genes Bax,Fas,Fas-L.The development of human uterine leiomyoma was relate to abnormal apoptosis.â‘¢The progesterone can not affect the expressions of apoptopic genes Bcl-2,Bax,Fas,Fas-L in uterine smooth muscle. |
