Objective To examine the expression of Lox-1 in aorta from cholesterol -fed rabbits and the effects of Losartan on atherosclerotic lesion and Lox-1. TO test weather Losartan have antiatherogenic effect.Methods Twenty-two rabbits were randomly divided into three groups: normal diet group(normal diet 100g/d), high-cholesterol group(high- cholesterol diet 100g/d), and Losartan group(high-cholesterol diet 100g/d+ Losartan power 25mg/d). The blood samples were collected for cholesterol and angiotensin II level at the begein of week O.week 6 and week 10. LOX-1 expression in aortas was examined by immunohistochemistry and semiquantitative RT-PCR. The ormation of aortic lesion were examined by stained in Herxheimers liquid.Results (l)Highcholesterol diet significantly high the level of blood TG TC(P<0.05) and angiotensin II (506.42 +87.59pg/ml vs 262.32 +36.01pg/ml), increased the ormation of aortic lesion (1.35 +0.06 vs 0.04 + 0.01) and the expression of LOX-1 mRNA (1.983 +0.020 vs 0.525 +0.019) and protain. (2)Treatment with losartan attenuated aortic intimal proliferation , decreased ormation of aortic lesion(0.25+0.06 vs 0.42+0.09) and the LOX-1 expression (1.309+0.018 vs 1.983 +0.020) . But the level of cholesterol did not alter significantly (P>0.05) . And the level of angiotensin II was much higher (789.50 + 94.08pg/ml vs 506.42 + 87.59pg/ml) .Conclusions (1 )High-cholesterol diet induces the expression of LOX-1 in neointima of aortas. Lox-1 have atherogenic effect. (2)Treatment with losartan, an ATI blocker, markedly decreases this enhanced LOX-1 expression. Losartan have antiatherogenic effect which connected with decreased LOX-1 expression. |
