| Objective To investigate the placental leptin rnRNA in normal pregnancies and patients with PIH and to study its clinical significance.Methods RT-PCR was used to measure the levels of leptin mRNA in 38 healthy normal the third trimester pregnant women and 62 patients with PIH (15mild, 18moderate and 29severe).Nitrite reductase was used to measure the concentration of nitric oxide of placenta . Placental leptin mRNA levels were correlated with NO,BP and albuminuria .Results 1. The levels of leptin mRNA of placenta in normal pregnant women and PIH were 0.139 0.12, 0.603 0.287 respectively .(The levels of placental leptin mRNA in mild, moderate and severe PIH were 0.145 0.056> 0.38 0.122> 0.752 0.199 respectively). 2. Leptin mRNA levels in PIH were higher than those in normal pregnant women ( P<0.01) and there was all obviously significant between mild .moderate and severe PIH (P<0.05), but there was no difference between mild PIH and normal pregnant women. 3. The concentration of placental NO in PIH group were lower than normal pregnant women (P<0.01 ) .there was great difference between mild, moderate and severe PIH (P<0.01 )0 4. leptin mRNA levels of placenta in PIH negatively correlated with NO concentrations of placenta in PIH (r=-0.6294,P<0.01 ). 5, The mRNA levels of placenta positively correlated with the levels of SBP, DBF, MAP and proteinuria (r=0.547, 0.377, 0.45, 0.517 ; P= 0.015, 0.015, 0.020 ,0.040 respectively) .Conclutions The resultes suggests that the levels of leptin mRNA in placenta are gradually increased .according to the mild, moderate and severe of PIH. Placental leptin probably reduce NO concentrations of placenta in PIH .Which might play an important role in pathogenesis of PIH. Elevated placenta leptin mRNA levels may be one of the monitor of PIH. |
