Pemphigus is a kind of severe disease in which skin and mucous membrane often get involved. It is clinically characterized by intraepidermis vesicles or bullas that are easy to break and positive Nikolskys sign. Pemphigus antibodies IgGs can be detected in the patients serum by indirect immunofluorescence.Corticosteroids are the main therapy for the treatment of pemphigus at present , but they have a lot of side - effects if they have been used for a long time. Examinating the deposits of pemphigus antibodies in skin has very important significance to the diagnosis of pemphigus and the treatment of patients. However, we cann' t check patients by biopsy repeatedly. Therefore, the purpose of the study is to develop a simple, fast and non - invasive method for the immuno-pathological diagnosis of pemphigus using plucked hair of the patients by direct immunoflurescence.Materials and Methods一 Materials;Hairs of Pemphigus patients; 21 patients from No. 1 Hospital of China Medical University are definitely diagnosed by clinical features and immunofluorescence. 40 hairs were plucked from each patient with pemphigus.8 patients with bullous pemphigoid and 10 healthy persons were used as controls.Major reagent: FITC - binding sheep - anti - human IgG antibody ( American SIGMA Corporation).二 MethodsHairs with follicles were stained by direct immunofluorescence.Results19 of 21 Pemphigus patients were positive with reticular deposits of IgG Pemphigus antibodies in the plucked hair follicles, 2 of 21 patients negative. Rticular deposits of IgG Pemphigus antibodies can be observed in the layers within the outer root sheath, but such pattern was not seen in healthy controls and bullous pemphigoid.ConclusionsThe examination of the deposits of IgG antibodies by DIF in the follicles of plucked hair from patients with pemphigus may be a convenient, sensitive, fast and non - invasive method for the diagnosis of Pemphigus. |
