As one appendix organ of skin, hair follicles play an important role in skin self- renewing hair growth and tumor origin. And deep burn wound healing mainly depends on the proliferation of appendix epithelia especially the hair follicle cells. In the clinical treatment, we usually find the outcome of burn wound is the scar formation and without hair regeneration. At the same time, wound healing of large area burn only depends on auto- or allo- skin graft because of absence of seed cells. The finding of hair follicle stem cells gives a promise for burn wound repairing rapidly and completely and artificial skin constructing. However, there are many problems about the hair follicle need to be clarifiediil) The factors affecting on hair growth and the control mechanism of hair cycle have not been definitely recognized. ()Sorne progress has been made in locating hair follicle stem cells, but not a breakthrough in isolating cultivating and identificating the stem cells in vitro.lthough possessing high proliferation ability, the outer root sheath cells(ORSc) showed difficult growth when cultured in vitro, which limited the research on the biology characters of ORSc.(41)The hair papilla cells are very important for the hair regeneration. Whether the hair papilla cells cultured could induce the hair regeneration?㏕here has not been an available drug that could effectively enhance the burn wound repaired completely including the formation of skin appendages such as hair follicle and sweat gland. Focusing on above questions we have done some work as follows.㏕he structure characters of scalp and hair follicle were studied in histology.㏕he cytokeratin expression was investigated by the immunohistochemistzy to analyze the proliferation ability of the epithelial cells in different region, and determine the location of stem cells. fter isolated from scalp tissue sample, the hair follicles as free organs were cultured in different mediums, to compare the mediums?effects on the hair follicle growth.㏕he hair follicle regeneration was observed through culturing the hair follicles transected at different site.()ORSc were cultured with hair follicle tissue or cells isolated by digestion, which has laid a foundation for exploring fwther the method of culturing hair follicle stem cells. e first described the growth curve cytokeratin expression~ cell ultrastruture and cell cycle of ORSc in detail, and compared them with normal keratinocyte's.()The hair papila cells were cultured, identiflcated, passaged and stored with liquid nitrogen, and their growth characters were observed. 甅inoxidil which has been used to treat hypertension or stimulate hair regeneration was investigated for its effects on the proliferation of ORSc and fibroblast. Our results as follows: (DA great number of hair folIicles sebaceous glands and sweat glands could be seen in the scalp. The hair follicle epithelia were fill of a great number of low differentiation cells. he in-tmunohistochemistiy of scalp showed the CK1 5 mainly expressed in the bulge of the hair follicle, and some of the extra layer of outer root sheath. CKI9 mainly expressed in the sweat gland~ the out layer of outer root sheath and the basal layer of skin.CK16 expressed both in the epithelia and the outer root sheath. CKIO mainly distributed in the suprabasal epithelia and the out layer of sebaceous gland. e cultured the whole hair follicles with different mediums.
