Objective:To study the expression of a novel inhibitor gene of apoptosis survivin,in bladder transitional cell carcinoma(TCC) and its relationship with the expression of bcl-2,p53,c-myc proteins.Methods:Expression of the survivin mRNA was evaluated by reverse transcriptase polymerase chain reaction(RT-PCR) in 37 bladder TCC tissues samples of it confirmed by pathology after operation and 12 normal bladder tissues samples. Immunohistochemical techiniques(SP) assay was to detect the expression of bcl-2,p53,c-myc proteins.Results:Normal bladder tissues did not express survivin. In contrast,survivin was expressed in 26 of 37(70.3%)of bladder TCC. Expression of survivin was related to the tumor grade and clinical stage. The positive rate of these three tumor grade was as follows:55.6%(gradeâ… ),72.7%(gradeâ…¡), 100%(grade â…¢)(â… +â…¡:â…¢,P<0.05) respectively, and the positive rate of clinical stage was 58.3%(stage Tis,a-1),92.3%(stage T2-4)(P<0.05). Bcl-2,p53,c-myc expression in bladder TCC(83.3%,20/24 versus 6 of 13 cases,P<0.05; 84.6%,22/26 versus 4 of 11 cases,P<0.05; 85.7%,18/21 versus 8 of 16 cases,P<0.05).Conclusions:Apoptosis inhibition by survivin,alone or cooperation with bcl-2,p53 and c-myc may participate in the onset and progression of bladder carcinoma.Survivin,participation of bcl-2, de-activation of antioncogene p53 and up-regulation of oncogence c-myc might play synergetic roles in the process of carcingenesis of bladder. |
