The Expression Of PPARγ In Nsclc And The Correlation Of PPARγ And Clinical Indexes

Objective To study the expression of peroxisome proliferator activated receptor γ(PPARγ)in non-small cell lung carcinoma(Nsclc) and PPARγwas correlated with tumor histological type ,grade,differentiation and metastasis of lymph nodes.To investigate the significance of PPARγ in Nsclc.Methods 60 pathological primary tumor specimens in random from surgerical patients with Nsclc during 1998 to 2002,none treated previously by chemotheraphy or radiation.40 patients were men and 20 were women with a mean age of 25-76 year.There were 33 cases of squamous cell lung carcinoma and 20 cases of lung adenocarcinoma.clinical stages(TNM):stage I30 cases,stageII 18 cases,stageIII-IV 12 cases.cell differentiation:high differentiation 22 cases,moderate differentiation 26 cases,low differentiation 12 cases.metastasis of lymph nodes 23 cases.21 normal lung tissues beside Nsclc were served as controls.Streptavidin-peroxidase(SP) and high pressure immunohistochemical methods were adopted to examine the PPARγ in 60 patients with Nsclc and 21 normal lung tissues.To analyze the relation of PPARγ and tumor histological type,stage,differentiation and metastasis of lymph nodes. Results Positive PPARγ expression was observed in 27 out of 60 cases of Nsclc examined(45%) and in 1 out of 21 normal lung tissues(4.76%).Therewas significant difference between two groups(P<0.01,respectively).Compared with those in high and moderate differentiation,expression of PPARγ of those in low differentiation showed an ascending tendency,There was a significant difference(P<0.01,respectively).Non-Statistically significant difference was observed for postive PPARγ expression between squamous cell lung carcinoma cases and adenocarcinoma ones(P>0.05).positive PPARγ expression was found in 12 out of 23 cases of metastasis of lymph nodes(52.17%) and in 15 out of 37 cases of no metastasis of lymph nodes(40.54%).non-statistically significant difference in positive PPARγ expression was found concerning the tumor stage .Conclusion High positive PPARγ expression was one of signals differentiating between Nsclc cases and normal lung tissues.PPARγ possibly participate the biological mechanisms in progression of Nsclc.PPARγ was inferred to correlate with the carcinogenic maliganant extent,possibly judging the prognosis.our results provided the important basis of applying PPARγ ligands to therapy lung cancer in future.
