Objective: To investigate the possibility of pathologic scars treated in endostatin , by observing the effects of endothelial cell and neogenesis capillary of pathologic scars treated in endostatin , through establishing models of hyperplastic scar in rabbit ear and injecting Endostatin . Methods: To total 32 rabbits, the wounds are created as follows: 06mm round wounds with total skin loses on the ventral side, 4 holes for each ear, summing total 256 holes. To divide 4 groups : Experiment groupl ( Endostatin treatment A) , Experiment group2 (Endostatin treatment B) Control groupl(triamcinolone acetonaide acetate control group) , Control group2(NS control group). The changes of 4 groups are evaluated through pathological sections HE staining , electron microscope and general observation. Results:!. General observation: The epithelization time of the wounds is in 15-20 days, 20 days after epithelization , the high of excess dermal scarring is as 3 times high as original ventol skin, color is pink, the blood vessel is abundant ; The scars begin to soften in about 60 days, the excess scarring can last nearly 100 days; The color and luster of scar after treating in Endostatin become light , the thickness is reduced, and become smooth. The area of scar in experiment groups is obviously less than control groups ( P <0. 01). 2. Pathological section : the counts of capillaries in experiment groups are obviously less than control groups ( P <0. 01) ; On the contrary, blood capillary and fibrocyte hyperplasia , the collagen deposits in a large amount. 3. Electronmicroscope:Treatment groups appear that endothelial cell wither and die, nuclear membrane pycnosis; Central groups are better than Treatment group. Conclusion: 1.To set up animal' s hyperplastic scar model by animal itself to the side of the belly of rabbit' s ear, it can produce excessive dermal scarring which is similar to human hypertrophic scar, it is significant for studying the formation and therapy of scar; 2. There are a large amount of endothelial cells and blood capillary hyperplasia in the formation of scar in rabbit ear, they verify that the formation of the blood capillary plays an important role in the formation of the hyperplastic scar;3. Endostatin could prevent and treat the scar through the endothelial cells apoptosis, suppressing the hyperplasia of endothelial cells and the formation of the new born blood vessel , offer a new way for treating the pathology scar clinically.
