Objective To explore the relationship between resistin gene polymorphism and type 2 diabetes mellitus in Chinese. The correlation between the A allele and HOMA-IR, waist-to-hip ratio, fasting plasma insulin was investigated as well.Methods The G/A phenotype at position 299 of resistin gene intron 2 was identified by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) in 186 type 2 diabetes patients and 61 healthy control subjects in Chinese. Total cholesterol, serum triglyceride and fasting plasma glucose were measured by CX7, Beckman and fasting plasma insulin by the Roche Elecsys 2010 analyzer, HbA1c by low pressure liquid chromatography analyzer. Alleles and genotypes were assessed by chi-square test, and continuous variables by analysis of variance and q test, correlations of multiple variables by logistic regression analysis.Results There was no difference in resistin gene polymorphism between type 2 diabetes mellitus and healthy control subjects. There was no difference in resistin gene polymorphism among three subgroups, classified by body mass index, of diabetic subjects. The clinical and biochemical data in different genotypes were analyzed in the group of type 2 diabetes mellitus. Fasting plasma insulin(10.46 7.90 vs 8.25 + 6.66 mlu/L, P<0.05; 14.11 + 8.60 vs 8.25 + 6.66 mlu/L, P<0.01; respectively), HOMA-IR( 4.32 + 3.04 vs 3.30 + 2.48, P<0.05; 6.47 + 3.71 vs 3.30 + 2.48, P<0.01; respectively), waist circumference(86.38 11.18 vs 82.86 9.52cm, P<0.05; 89.25 10.79 vs 82.86 + 9.52cm, P<0.05; respectively) and waist-to-hip ratio( 0.86 + 0.07 vs 0.83+0.06, P<0.05; 0.89 + 0.07 vs 0.83 0.06, P<0.01; respectively) of the GA and AA subjects were significantly higher than those of the GG subjects. Fasting plasma insulin, HOMA-IR and waist-to-hip ratio were increased significantly in the AA subjects as compared with those in the GA subjects. Logistic regression analysis showed that waist-to-hip ratio and HOMA-IR were associated to the A allele independently.Conelusion There is no relationship between resistin gene polymorphism and type 2 diabetes mellitus in Chinese. The A allele in resistin gene intron 2 probably contributesto central obesity and obesity-associated insulin resistance in type 2 diabetes mellitus patients. |
