Prostate cancer is a common malignant tumor in men. Molecular study indicates that the origination of prostate cancer is related to the increment of cell proliferation and reduction of apoptosis. Today, experiments have proved that apoptosis-associated factor Fas and TRAIL as well as GnRH are expressed by this cancer and the extents of expressions are different from normal tissues significantly, these indicate that Fas, TRAIL, GnRH have some relationships with the production and development of prostate cancer. However until now there are few works on the relationships of these factors.To further discuss the relationships among Fas, TRAIL and GnRH, prostate cancer cell line PC3 and prostate cancer tissue were used in this experiments, as well as immunohistochemistry, cell culture. Flow CytoMeter, image analysis technique were applied to achieve our aim.The experiments results as follow:l.Alarelin effected the expression of Fas on PC3 cell topeak at day 4 when the concentration of Alarelin was 1.0 x10-9mol/L, but this effection was rather short and weak.2. Prostate cancer tissue (n=19) all showed GnRHR and Fas immuno-reactivity, immuno-positive material mainly distributed in cell membrane and cytoplasm, karyon was negative. In prostate cancer tissue with higher differentiation, the reaction of GnRHR immunoreactive cells was stronger and the cell was dyed deeper. However prostate cancer tissue with lower differentiation had reverse outcome. In prostate hyperplastic tissue, the reaction of GnRHR was negative. From another hand, in prostate cancer tissue with higher differentiation, the reaction of Fas immunoreactive cells was feebleness and the cell was dyed slighter. However prostate cancer tissue with lower differentiation had reverse outcome. In prostate hyperplastic tissue, the reaction of Fas was also positive. These mean that the current of GnRHR variety is opposite to Fas in prostate cancer.3. Prostate cancer tissue (n=20) all showed TRAIL immunoreactivity; immuno-positive material mainly distributed in cell membrane and cytoplasm. In prostate cancer tissue with higher differentiation, the reaction of GnRHR immunoreactive cells was stronger and the cell was dyed deeper. However prostate cancer tissue with lower differentiation had reverse outcome. In prostate hyperplastic tissue, the reaction of Fas was also positive, but the reaction was weaker than that in prostate cancer tissue. These mean that TRAIL has the same variation currentas GnRHR in prostate cancer.Conclusion:1. Alarelin may effect the expression of Fas on PC3 cell slightly and shortly.2.There are expressions of GnRHR Fas and TRAIL in prostate cancer with different proliferation. The expression tendency of GnRHR is consistent with that of TRAIL, but is contrary to that of Fas.
